Tesamorelin: When I Increased My Dose… the Scale Jumped 8 lbs 😳
Personal Experience Update: I know we already did a deep dive on Tesamorelin, so I wanted to share my personal experience with it so far, because this was a good reminder that protocols don’t always play out the same way for everyone. I started at 250mcg and tolerated it really well the first week. Since everything seemed good, I increased it to 500mcg. Within that week I noticed pretty significant water retention and the scale went up almost 8 pounds. Now logically I knew there was no possible way to gain that much fat that fast — but seeing the number jump like that definitely messes with your head a little. I decided to stick it out another week to see if my body would adjust… but the water retention didn’t change. So I dropped back down to 250mcg, which is where I’m currently at, and the water weight has already started coming back down again. It was honestly surprising how just a small dose increase made such a big difference. Some people’s bodies are just more sensitive to certain peptides, and I think I’m probably one of those people. My protocol originally had me titrating up to 1mg by June 16, but this experience reminded me that more isn’t always better. Sometimes the smartest move is listening to your body and adjusting, even if that means staying at a lower dose or moving up more slowly. Curious if anyone else here has had a similar experience with Tesamorelin — especially when increasing doses. Did your body eventually adjust or did lowering the dose help?

I am would love to try this one but so concerned about the water retention and joint pain. Any issues with pain? Also wondering if stacking with KPV would be beneficial for the water retention? Thanks for sharing and excited to see how you go with this one! It is on my list that I am really curious about.

Labs and Hormones checked 1st!!
I feel like a lot of people get this backwards when it comes to peptides and ā€œbiohacking.ā€ So many people jump straight into stacking things without first making sure the foundation is dialed in. For me, one of the biggest things was getting bloodwork done and optimizing my hormones. I actually started testosterone pellets back in 2023 through a local provider and saw a huge benefit just from that alone — my energy improved a lot. The only downside with pellets is they can feel a little inconsistent and they’re hard to adjust once they’re inserted. But it allowed me to also get my lab work done every 12 weeks and check my hormone levels. Just this week I met with Defy Medical after seeing a lot of people in another group I’m part of mention them for going through labs and hormone optimization and testosterone injections which I think may be more consistent and allow adjustments if needed. They review everything with you — hormones, testosterone levels, and anything else that might need attention. My initial consult was $325, they ordered labs, and I just took the lab order to Quest and ran it through my insurance. What was really interesting is that these labs were done about 3–4 months after my last labs and since then I had started peptides, and a lot of my markers had actually improved. I’ll be honest, I was a little nervous to see the results because even though I feel great, you always wonder if your bloodwork is going to reflect that too. Thankfully it did, which was really reassuring. It was a good reminder that sometimes it’s not about adding a bunch of random things — it’s about understanding your body and filling in the gaps where needed. Once that foundation is there, then things like peptides can become another tool to support your goals. Just sharing what I’m personally doing and what I’m learning along the way. Any chance I get to talk about the wonders of HRT and how it can help - I will! Around 50 (yep) I was finally taken off birth control as I no longer needed it. What I did not know is how it was balancing my hormones. Within a month I was miserable. Moody, hot flashes, generally just felt horrible. I tried every supplement I could find and ended up finding Mary Claire Haver, MD on Instagram and the benefit of HRT. I have been on HRT for a few years now and it has been amazing. There are so many resources where you can find support now! Peptides are a game changer, but if you are close to 40 - consider balancing hormones first then peptides. Just my experience - and I hope it reaches someone and it helps.

Hi! I have been on GLP2 (Tirzepitide) since December of 2024. I started with a medical clinic, then moved to Ellie MD and now I am researching with research grade. I have also researched a few other peptides to support stress management, nerve pain from 4 back surgeries, inflammation and other perimenopause symptoms.
