SS-31 Mitochondrial Restoration
Once the mitochondrial assessment points toward impaired mitochondrial function, the objective changes from optimization to restoration.
This is where SS-31 (Elamipretide) comes into the picture.
🔧 SS-31 — THE ENGINE REPAIR KIT
SS-31 is a mitochondrial-targeted peptide with an affinity for the inner mitochondrial membrane—the highly folded internal membrane where the electron transport chain operates and most cellular ATP is produced.
Central to that membrane is cardiolipin.
Cardiolipin is a unique and essential phospholipid found almost exclusively within the inner mitochondrial membrane, where it accounts for roughly 20% of its lipid composition. It is often referred to as the “signature lipid” of mitochondria because of the important role it plays in mitochondrial structure and cellular energy production.
Cardiolipin helps organize and stabilize the inner mitochondrial membrane and supports the protein complexes that make up the electron transport chain—the machinery responsible for producing most of our ATP.
When cardiolipin becomes damaged or oxidized, that organization can begin to break down. Electron transport becomes less efficient, oxidative stress can increase, and the mitochondria may become less efficient at converting nutrients and oxygen into usable cellular energy.
This is the environment SS-31 targets.
SS-31 interacts with cardiolipin at the inner mitochondrial membrane. Research suggests this interaction can help stabilize the membrane environment, support the organization and function of the respiratory machinery, improve mitochondrial bioenergetics and reduce excessive mitochondrial oxidative stress.
The potential result is a mitochondrial environment better equipped to do what it was designed to do:
Produce ATP efficiently.

⚙️ THE 8-WEEK SS-31 RESTORATION PROTOCOL
The restoration phase is structured around 8 weeks of consistent daily SS-31 administration.
Duration: 8 weeks
Frequency: Daily
Working range: 5–10 mg per day
Timing: Same administration time every day - Consistency is an important part of this playbook.
Choose an administration time that can realistically be maintained throughout the entire eight weeks and stay with it. If the selected time is 7:00 AM, the objective is to administer SS-31 at approximately 7:00 AM every day—not early morning one day, lunchtime the next and evening the day after that.
Pick the time. Establish the routine. Keep it consistent for eight weeks.
SS-31 is increasingly being seen in 30 mg and 50 mg vial strengths, whereas 10 mg vials were once much more common. The larger vial strengths simply make a multi-week daily protocol more practical.
📊 TRACK THE RESTORATION PHASE
Throughout the eight weeks, continue tracking the same core indicators established during the self-assessment:
Energy • Endurance • Exercise Performance • Recovery • Resting Heart Rate • Sleep
The emphasis is on trends rather than individual days.
One great workout doesn't establish improvement. One poor night's sleep doesn't establish failure.
What we're looking for is whether a consistent pattern begins to develop over the eight-week period.
More stable daily energy, greater exercise tolerance, improved recovery, changes in resting heart rate or better sleep become considerably more meaningful when they persist.
And if very little changes, that is meaningful information as well.

At the completion of Week 8, return to the same self-assessment used before beginning the protocol.
Compare where you finished with where you started:

The objective isn't to complete eight weeks and simply declare the mitochondria “repaired.”
It's to determine whether the restoration phase produced a meaningful and sustained change in mitochondrial function. That's why establishing the starting point mattered.
